One of the major verses that we can use to explain the Imamate of Imam Ali (AS) is by using the verse of Wilayat, Surah Maidah, Verse 5. It is stated as follows
إِنَّمَا وَلِيُّكُمُ اللّهُ وَرَسُولُهُ وَالَّذِينَ آمَنُواْ الَّذِينَ يُقِيمُونَ الصَّلاَةَ وَيُؤْتُونَ الزَّكَاةَ وَهُمْ رَاكِعُونَ ﴿٥٥﴾
(5/55) Your guardian is only Allah, His Apostle, and the faithful who maintain the prayer and give the zakat while bowing down.
